I'll take a stab at this- M1918 Browning Automatic Rifle. The 1903 rifle was DESIGNED for the 30-03. The 1917 Enfield was DESIGNED for the .303 Enfield. The BAR was the first DESIGNED to use the 30-06.

C3, I was incorrect in stating your answer, the BAR, was correct, so the question as to what weapon originally designed for the 30-06 cartridge was adopted by the U.S. military, still stands.

Trez, if you mean the Browning Model 1917 machine gun, you are correct, in that it was the first weapon, originally designed for the 30-06 cartridge, to be adopted by the U.S. military.
